Offer Description

The contract funded by the “Maison de la Chimie” Foundation will be a continuation of the ANR SCHIZOLIP project, which focuses on the role of the membrane lipid environment on the heteromerization of the dopamine D2 G-protein-coupled receptor (GPCR) with other GPCRs. The project will involve the production, purification, and characterization of the receptors of interest, in the presence or absence of their effectors (G protein, β-arrestin). The objective is to determine the structure of protein complexes formed in the presence of different surrounding lipids using cryogenic electron microscopy (cryoEM). To achieve this, various molecules can be used to "extract" the receptors from the membranes: copolymers, detergents, etc.

- Cell culture (Sf9 insect cells, HEK mammalian cells)
- Protein production, purification, and characterization
- Biophysical methods for characterizing membrane proteins
- Cryogenic electron microscopy (cryoEM)
- Analysis, formatting, and presentation of results
- Oral presentations in English

The project will be carried out under the supervision of Estelle Rascol at the Institute of Chemistry and Biology of Membranes and Nano-Objects (CBMN) at the University of Bordeaux, in collaboration with members of Isabel Alves' team and the project partners. The laboratory is equipped with most of the instruments needed to carry out the project.

- Knowledge of G-protein-coupled receptors
- Good knowledge of biophysical techniques applied to the study of proteins
- Expertise in membrane protein biochemistry
- Experience in cryoEM is a plus
- Good command of English (oral and written)
- Writing and oral communication skills
- Teamwork
